Description: | Induction of P57 reduced proliferation of HepG2 and SNU398 HCC cells, as demonstrated by growth curve analysis.FACS analysis revealed that P57 transfected cells accumulate in G1 phase of cell cycle. Seventy-two hours post-P57 transfection, cells began to change the morphology, becoming enlarged and flattened, and they adopted a senescent phenotype when compared to vector-only cells .In accordance with the senescence phenotype, induction of P57 in HepG2 and SNU398 led to the identification SA-β-gal positive cells.An increase in P16 protein levels was also observed in HepG2 cells. |

Pathway description: | Notch3-and Notch1-silenced cells showed a down-regulation of Hes1 target gene together with an up regulation of P57 mRNA and protein levels .Down-regulation of Hes1 significantly increased P57 mRNA and protein levels in both analyzed cell lines .In HepG2 cells, but not in HepG2, with siRNA to Hes1, DNA of the P57 promoter region could be specifically detected in the Hes1-immunoprecipitated DNA complex from formaldehyde-treated cells, indicating Hes1 occupancy at the P57 promoter in vivo. |
