Experiment: SA-β-gal activity assay//Flow cytometry
Description: In the control cells, only 9.3 ± 1.9% cells were positive for SA-β-gal staining but, after treatment with H2O2, the percentage of SA-β-gal-positive cells increased to 63.5 ± 5.8% at 100 μM, 86.4 ± 5.4% at 200 μM, and 93.4 ± 7.3% at 400 μM. Furthermore, the cell cycle phase distribution was examined. Cells treated with 100 μM and 200 μM H2O2 exhibited a significantly increased proportion in the G0/G1 phase (by 11.1% and 11.9% in comparison to the control, respectively). Meanwhile, H2O2-treated cells showed a lower percentage in the S phase (10.2 ± 0.9% in control cells, 3.5 ± 1.3% at 100 μM, and 3.7 ± 0.2% at 200 μM).
