Description: | We found that treated cells had dramatic increases in both SAβGAL and Lysotracker staining ,compared to the controls (DMSO). These changes are indicative of lysosomal stress and induction of senescence.Notably, we also observed a statistically significant increase in mean nuclear area from 200 lm2 in control cells to 245 lm2 which occurred after only 72 h TnV6 treatment.We observed a dramatically increased and highly reticular mitochondrial load following TnV6 treatment, compared to control proliferating fibroblasts (DMSO alone) which exhibited a low mitochondrial load, with the majority of cells showing a perinuclear distribution of mitochondria.A marked increase in senescent-like actin stress fibres was observed in cells treated with TnV6 compared with vehicle-only control cells, which showed more diffuse actin patterns characteristic of proliferating cells . |
