Description: Metformin inhibited replicative senescence in WI‐38 and IMR‐90 cells. Treatment of cells with metformin delayed IR‐induced senescence, as assessed initially by SA‐β‐gal‐positive cells and subsequently confirmed by examination of two well‐established protein markers of senescence, p16 and p21. All of these markers of senescence were decreased by metformin treatment.

Target gene description: Livers of metformin‐treated mice had higher DICER1 protein levels than control mice .DICER1 mRNA levels further increased by metformin treatment, which may reflect an effect on DICER1 at the transcriptional level. SiRNA‐mediated downregulation of DICER1 levels increased cellular senescence and blocked the ability of metformin to lower the number of SA‐β‐gal‐positive cells in both WI‐38 and IMR‐90 cells. Conversely, overexpression of DICER1 decreased cellular senescence .
