Description: | Inhibition of proliferation was confirmed by cell staining with antibodies to Ki-67, a marker of proliferative activity. Seventy-two hours after heat shock, only a few cells were Ki-67-positive. Cell cycle analysis by flow cytometry revealed that heated cells were arrested mostly in G2/M phase.The modified morphology of MSCs subjected to HS: they are enlarged,more flattened, and express X-gal. Three days after HS, the number of SA-β-gal-positive cells increased to about 70 %. HS induced expression of p21 protein, a cell cycle inhibitor and major regulator of the senescence program. |
