Description: | After treatment with 0.5 μM RD, accumulation of cells with enlarged and flattened morphology was evident in PCa cells. The level of SASP mark IL-6 was found significantly increased in PCa cells after RD treatment by Elisa assay and q-PCR assay . In agreement with a large morphology, increased SA-β-gal was also observed in response to RD . The SA-β-gal positive stained cells was significantly enhanced by LNCaP cells,DU145 cells, respectively, as compared to controls. |

Target gene description: | Deletion of p21CIP1 by targeting siRNA impaired RD-induced senescent (SA-β-Gal positive) cells in all three PCa cells.SA-summ staining was decreased 0.32±0.05-fold in LNCaP, 0.43±0.19-fold in PC3, and 0.31±0.03-fold in DU145 cells, respectively, indicating that RD-induced cellular senescence was, at least in part, in a p21CIP1-dependent manner. |
