Description: | Using the Telomeric Repeat Amplification Protocol to measure telomerase activity in monolayer and mammosphere cells, we observed that both cell types displayed telomerase activity, which increased after irradiation with 1 and 10 Gy. the telomerase activity in the mammospheres was statistically significantly more robust than in the monolayer population .and indeed analysis of senescence based on cellular β-galactosidase activity revealed that the percentage of senescent mammospheres is approximately half that of the monolayer population both prior to irradiation and three days after irradiation with 4 Gy. |

Target gene description: | An examination of other key proteins involved in the regulation of senescence revealed that the mammosphere cells, in comparison to the monolayer cell population, expressed markedly lower levels of the senescence signaling protein, p21, especially in unirradiated cells. In the case of pRb there was a notable reduction in total protein following irradiation with 1 Gy and also a lower degree of phosphorylation after 10-Gy irradiation of the mammospheres. |
