Aging Overview

External factors

External factors: Nutlin-3
Aging type: Accelerate
Aging characteristic:
Category: Chemical compounds
Phenotype: Sarcoma
Experimental category: L
Tissue type: --
Cell name: CADO-ES-1
PMID: 21334198
Experiment: SA-β-gal activity assay
Description: After a 4-d exposure to nutlin-3 at a dose (2 lM) that had hardly exerted any apoptosis-inducing effect, CADO-ES-1 cells were stained for SA-β-Gal activity, the most widely used marker for cellular senescence. Nutlin-3-treated cells showed clear signs of senescence, whereas untreated cells did not.

Regulatory relationship

Target gene: P53
R-EF-Target gene: Activation
Official symbol(s): P53
Target gene experiment: Western blot//qRT-PCR
Target gene description: Immunoblot detection revealed a rise of p53 after nutlin-3 exposure in the ES cell lines with wt-p53.To confirm the p53-activating effect of nutlin-3, we determined the gene expression of three key transcriptional targets of p5326 MDM2, p21 and PUMA by real-time RT-PCR.Nutlin-3 induced concentration-dependent increases in mRNA levels in all three cases in the wt-p53 cell lines, but not in the mt-p53 one.
