Description: | Indeed, after 5 days of treatment in vitro,we observed that the cellular size was greatly enlarged ,which is a characteristic associated with senescence.β-galactosidase activity was evaluated and found to be enhanced in drug-treated Hs294T cells and in other melanoma cell lines (SK-Mel-2, SK-Mel-5 and SK-Mel-28).The results demonstrated that IL-6, IL-7, IL-10, GM-CSF, IL-8, RANTES, GRO and GRO-a were upregulated in response to drug treatment. |

Pathway description: | We knocked down either ATM or Chk2 and found that senescence was reduced >30% in knockdown cells,indicating that the ATM/Chk2 pathway mediates MLN8237-induced senescence.An NF-kB luciferase assay was also performed using Hs294T NF-kB reporter cells. NF-kB transcriptional activity was significantly increased after treatment with 1mM MLN8237 for 5 days . |
