Pathway description: | Western blot:Since Sjp40 treatment markedly increased the protein level of P27。SA-β-gal activity assay//Knockdown:SA-β-Gal positive percent mediated by Sjp40 in LX-2 cells obviously reduced via knockdown of P27, which confirmed the crucial role of P27 in regulating cellular aging induced by Sjp40。Western blot:SKP2 over-expression reversed P27 protein level induced by Sjp40, indicating that P27 expression induced by Sjp40 is dependent on SKP2 in LX-2 cells.SA-β-gal activity assay:Over-expression of SKP2 rescued the Sjp40-induced senescence analyzed by SA-β-Gal assay. |
