External factors: | Baicalin |
Aging type: | Accelerate |
Aging characteristic: |
Category: | Chemical compounds |
Phenotype: | Colorectal cancer |
Experiment: | CCK-8 assay//SA-β-gal activity assay//Colony formation assay |
Description: | CCK-8 assay: In CCK-8 assay, baicalin inhibited the viability of HCT116 and SW480 colon cancer cells.Treatment with baicalin at concentrations of 10–40?μM led to significant increase of the percentages of SA-β-gal-positive cells with a dose-dependent manner.SA-β-gal activity assay:Treatment with baicalin at concentrations of 10–40?μM led to significant increase of the percentages of SA-β-gal-positive cells with a dose-dependent manner.Colony formation assay: In a colony formation assay, baicalin treatment significantly reduced the numbers of colonies in colon cancer cells at all doses tested. |
Target gene: | DEPP |
R-EF-Target gene: | Upregulation |
Official symbol(s): | DEPP1 |
Target gene experiment: | Western blot//qRT-PCR |
Target gene description: | Western blot//qRT-PCR:To further explore the regulation of DEPP in a hypoxic condition of tumor cells, HCT116 cells were placed into a hypoxic incubator maintained 1% O2. Western blotting and qRT–PCR analyses showed that the expression of DEPP was significantly upregulated . |
Regulatory pathway: | Ras-Raf-MEK-ERK//p16-Rb |
R-EF-Pathway: | Activation//Activation |
Official symbol(s): | ZHX2-MEK-ERK//CDKN2A-RB1 |
Pathway experiment: | Western blot |
Pathway description: | Western blot: In addition to the increased expression of DEPP, the phosphorylation and the activation of Raf1 and ERK were dramatically induced in the colon cancer cells in response to the hypoxic condition. Western blot: we found that the administration of baicalin in HCT116 cells only significantly affected the protein levels of p16INK4A and Rb during the induction of cellular senescence.Western blot: Similar results of DEPP upregulation and the activation of Ras/Raf/MEK/ERK and p16INK4A/Rb pathways were also observed in A549 and Panc-1 cell lines. |
