Description: Similar to silencing of TWIST1, treatment of the KRAS mutant NSCLC lines A549, H460, and H358 with harmine-induced changes characteristic of OIS, including positive Senescence-Associated β-galactosidase (SA-β-Gal) staining and induction of p21 and p27. We also found that harmine treatment induced OIS in NSCLC cell lines with EGFR and MET mutations .

Target gene description: Harmine treatment reduced the levels of TWIST1 protein in a dose- and time-dependent manner as shown by Western blotting. Remarkably, treatment with harmine resulted in a dose-dependent decrease of E2A protein expression in both KRAS-mutant and MET-mutant/amplified NSCLC cell lines.
