Description: | We tested senescence markers in LMNA-mutated and observed that SA-β-galactosidase activity was increased by 2.2- to 2.5-fold in LMNA mutant versus WT.The protein expression of the senescence markers, p16INK4, p21WAF, p53 and phospho-p53 was also altered. LMNA mutant harbored the typical morphology of senescent cells, with an increased number of prelamin A-stained nuclei showing dysmorphies, and of foci containing phosphorylated histone γ-H2AX indicating DNA damages. |

Target gene description: | We first evaluated the expression of miR-141-3p, which was shown by Yu et al. to regulate ZMPSTE24 protein expression during cellular senescence [40]. The level of miR-141-3p was increased by 1.5-fold in VSMCs expressing D47Y and R482W LMNA mutant and by 2-fold in LPV/r-treated cells, as compared to control cells. |
