Experiment: | TRAP assay//Flow cytometry//SA-β-gal activity assay |
Description: | Using the TRAP assay, we found that both nTregs and iTregs could induce the inhibition of telomerase activity in all the target cells.Upon co-cultivation with Tregs, we observed a significant decrease in the number of target cells in the S and G2/M phases and a reliable increase in the number of cells in the G0/G1 phases 30 days after transfection. β–Gal expression levels remained stably high over the days 60. β–Gal enzymatic activity also increased 45 days after transfection and remained high on day 60. |
Target gene: | α+β+HTERT |
R-EF-Target gene: | Upregulation |
Official symbol(s): | α+β+HTERT |
Target gene experiment: | Western blot//RT-PCR |
Target gene description: | Western blotting results showed a significant decrease in full-length α+ β+ hTERT and an increase in α+β– hTERT and EndoG in the target cells co-cultured with nTregs or iTregs Real-time RT-PCR revealed that cultivation of the target cells in the absence of iTregs resulted in a gradual increase in α+ β+ hTERT expression, decrease in α+ β– hTERT expression and decrease in EndoG expression to the levels of the control target cells over 48 h. |
