Description: Selective inhibitor of CXCR2, SB225002, was confirmed to inhibit the proliferation of lung cancer cells in a both time-dependent and dose-dependent manner. To investigate the role of CXCR2 in tumor cell senescence, the expres- sion of SA-β-gal was detected while using SB225002 on LL2 and H460 cells. After 24 h, the result showed that the percentage of β-gal-positive cells in SB225002 group was higher than that of control group. The senescence-associated markers p16 and p21 were up- regulated after using SB225002 and down-regulated after using CXCL2.

Pathway description: After using SB225002 and CXCL2 on LL2 cells, p38/ERK MAPK signaling pathway was found involved in CXCR2-associated signaling pathway. The levels of p38 and ERK proteins phosphorylation were obviously al- tered, whereas p-JNK almost remained unchanged during activation or inhibition of CXCR2.
