Description: | In our experiment, Gadd45α was down-regulated along with p53 in 293T cells transfected with both miR-25 and miR-30d and a cell cycle analysis showed that fewer cells were arrested at the G2-M phase than in the control. We then performed a cell cycle analysis and found that fewer cells were arrested at the G1-S phase (G1 arrest) when miR-25 or miR-30d was co-overexpressed with the p53 gene with a wt 3’UTR but not with the one with a respective mutant.Senescence was reduced in HCT116 cells with miRNA overexpression and lower levels of p53 and p21. |

Target gene description: | We found that both miR-25 and miR-30d down-regulated the expression of the ectopic p53 gene carrying a wt 3’UTR but not the one with a mutant. The expression of both p21 and Bax was either reduced or minimally changed, following p53 expression. These results suggest that miR-25 and miR-30d regulate cellular senescence through down-regulating p53 expression and subsequently the expression of p21 and that such regulation depends on miRNA binding sites of TP53 3’UTR. |
