Category: | ncRNA |
Phenotype: | Hepatocellular carcinoma |
Experimental category: | L |
PMID: | 28258280 |
Experiment: | SA-β-gal activity assay//BrdU assay |
Description: | Telomeric repeat amplification protocol (TRAP) assay, senescence-associated β-galactosidase assay and BrdU incorporation assay were performed 24 and 48 h after transfection. The results showed that miR-138 functioned to decrease telomerase activity, which in turn induced cell senescence and suppressed cell proliferation. |
Target gene: | TERT |
Official symbol(s): | TERT |
R-AG-Target gene: | Downregulation |
Subcategory: | Unclear |
Target gene experiment: | Western blot//qRT-PCR//Luciferase reporter assay |
Target gene description: | The binding site of miR-138 in the 3’UTR of TERT gene was predicted by miRNA target prediction tools. The results showed that wild-type miR-138, but not mutant miR-138 (mutated in the seed-complementary sequence),can significantly suppress the luciferase activities of Huh7 cells transfected with wild-type TERT 3’UTR construct,Quantitative real-time PCR and Western blotting analysis showed that wild-type miR-138 mimic can suppress endogenous TERT mRNA and protein expression in Huh7 and HepG2 cells. These results indicated that TERT can be a direct target of miR-138 in HCC cells. |
