Description: In the miR-141-3p-transfected cells, MTT assays and cell cycle analyses revealed that the growth rate was reduced 48?hours post-transfection. In contrast, the transfection of hMSCs with anti-miR-141-3p yielded an increase in proliferation. The correlation between the expression of ZMPSTE24 and miR-141-3p was confirmed in five other hMSC cell lines .The overexpression of miR-141-3p also increased SA-β-gal activity and γH2Ax expression in these cells.

Target gene description: We further investigated whether miR-141-3p regulates ZMPSTE24 expression, prelamin A accumulation and nuclear abnormalities. The transfection of hMSCs with miR-141-3p decreased ZMPSTE24 expression, resulting in increased expression of prelamin A, p16INK4A?and γH2AX. Conversely, these effects were reversed by miR-141-3p knockdown.
