Gene name: | CTSL |
Aging type: | Accelerate |
Aging characteristic: |
Tissue type: | -- |
Cell name: | MEF |
Experiment: | Western blot |
Description: | Altogether, our data demonstrate that the upregulation of CTSL activity upon loss of A-type lamins leads to degradation of 53BP1 and defective repair of DNA DSBs by NHEJ.Whereas Lmna-/-fibroblasts presented defects in the fast phase of repair corresponding to NHEJ, depletion of CTSL was sufficient to restore the repair of IR-induced DSBs to a degree that was indistinguishable from that of WT cells. |
Target gene: | A-TYPE LAMINS//53BP1//NHEJ |
Official symbol(s): | A-TYPE LAMINS//TP53BP1//NHEJ |
R-AG-Target gene: | --//Downregulation//-- |
Subcategory: | Unclear |
Target gene experiment: | Western blot |
Target gene description: | Western blots showing that acute depletion of CTSL by lentiviral transduction with an shRNA leads to stabilization of 53BP1 protein in Lmna-/-MEFs.CTSL overexpression led to increased levels of the Activate form of the enzyme and increased CTSL activity, as well as a decrease in 53BP1 levels.Altogether, our data demonstrate that the upregulation of CTSL activity upon loss of A-type lamins leads to degradation of 53BP1 and defective repair of DNA DSBs by NHEJ. |
