Description: | Senescence-associated (SA) β-galactosidase positive cells were up to 48.0 and 77.6% of the detected HCT116 p53 wt and HCT116 p53-/- cells, respectively. Using real-time qPCR, we examined the expression 12 senescencerelated genes, and found that in HCT116 cells p15, p21, and Rb were also upregulated in addition to p16.We further examined the senescence associated secretory phenotype (SASP) in MCF7, HCT116 and HCT116 p532/2 cells by real-time qRTPCR.IL-6, IL-8, IL-18, IL-1b, TNF-aCXCL1 and CXCR2 were all greatly induced, suggesting that the p65/S536D indeed has the function of wild type RelA/p65 phosphorylation at Ser536 in regulation of SASP phenotype. |
