Aging Overview

Aging gene

Gene name: TOP1
Aging type: Prevent
Aging characteristic:
Tissue type: --
Cell name: HCT116
Gene ID: 7150
Category: protein coding
Phenotype: Aging
Experimental category: L
PMID: 20682043
Experiment: SA-β-gal activity assay//Knockdown//Flow cytometry//Clonogenic assay
Description: We observed using clonogenic assays that sn38 induced a complete inhibition of cell proliferation. Using beta-galactosidase staining, we also noticed an induction of senescence following genotoxic treatment.control cells were synchronized in the G2 phase of the cell cycle after 48-72 hr.

Regulatory relationship

Target gene: AURORA-A
Official symbol(s): AURORA-A
R-AG-Target gene: --
Subcategory: Unclear
Target gene experiment: qRT-PCR//Western blot//Knockdown
Target gene description: Although this was the cell cycle stage when Aurora-A expression was supposed to be maximal, results indicated that the expression of the kinase was downregulated in response to sn38, both at the protein and mRNA levels. Finally, these experiments have also been repeated in a different colorectal cell line and sn38 also downregulated Aurora-A in HT29 cells.
