Description: | In the differentiated myo-tube from (P)RR‐expressing C2C12 cells, we detected positive SA‐β‐gal staining with an increase in γH2AX, a senescent marker, and increased expression of TNF‐α, a senescence‐associated secretory phenotype, indicating that (P)RR expression induced cellular senescence. Moreover, aging markers, such as p16, p21, and γH2AX, were up‐regulated in the muscles of both (P)RR‐Tg mice and aged WT mice. |

Pathway description: | As expected, ABC was significantly increased and nuclear localization of β‐catenin could be observed in the muscles of (P)RR‐Tg mice, indicating activation of Wnt/β‐catenin signaling by (P)RR .Wnt and YAP activities were enhanced in myotubes from (P)RR‐expressing C2C12 cells. Treatment with verteporfin, a small molecule that inhibits the assembly of YAP/TEAD and its transcriptional activity, significantly restored myotube formation in (P)RR‐expressing myoblasts. |
