Description: | Significant increases in SA-β-Gal activity were seen in all 3 lines when stably transduced with shSOX10 hairpins, suggesting that loss of SOX10 induces senescence in melanoma cells. Another hallmark of cellular senescence, senescence-associated heterochromatin foci (SAHF), was observed upon chromatin binding protein HP1b immunostaining .One week after infection, the shSOX10 lines showed reduced SOX10 protein levels and the cells became enlarged, flat, and translucent ). These cells also showed arrested or slowed growth. |

Target gene description: | In all three melanoma lines, SOX10 knockdown led to reduced levels of E2F1 protein and total RB protein.Reduced MITF protein was observed upon shSOX10 knockdown in both 0380-MMU and 0002-ARM cells.We observed moderate p16 expression in 0380-MMU cells;however,p16 levels remained unchanged following SOX10 knockdown. Weconsistently observed increases in p27 levels in all three cell lines as a result of SOX10 knockdown, similar to previous MITF knockdown studies. However, SOX10 knockdown caused an increase in p21 protein in all three-cell lines, thus showing a distinct difference from the MITF knockdown results. |
