Description: PRMT6 KD in both cell lines inhibits growth and specifically leads to a reduction of cells in S phase.Both sh-1 and sh-2 cells show a clear change in morphology, with cells looking bigger and flatter as compared with the scrambled control shRNA (sh-c) . As the morphology and the cell cycle arrest was reminiscent of cellular senescence, we confirmed this by SABG on the PRMT6 KD and control cells.

Target gene description: Although p16 levels did not change in either MCF7 [the p16 locus is deleted in these cells (24)] or MDA-MB-231 (wt p16), we observed a striking upregulation of p21 messenger RNA (mRNA) and protein levels. In MCF7 cells, the p21 promoter is bound by PRMT6, methylated on H3R2me2a and H3K27me3, and p21 is expressed at low levels.
