Description: | We found that upon withdrawal of WRN, the morphology of untransformed primary fibroblasts changed progressively within 5 days posttransfection. The cells became enlarged, flattened and developed the SA-β-Gal activity with a concomitant moderate decline of cumulative cell number.The significant hypophosphorylation of Rb was accompanied by decreased proliferation of the WRN-deficient cells, as evidenced by an average 40% reduction of bromodeoxyuridine incorporation (data not shown) and a similar decrease in S-phase cells with an increased proportion of G0-G1 cells by FACS analysis. |
