Aging Overview

Aging gene

Gene name: AKT1
Aging type: Accelerate
Aging characteristic:
Tissue type: --
Cell name: BJ-T
Gene ID: 207
Category: protein coding
Phenotype: Aging
Experimental category: L
PMID: 21909130
Experiment: SA-β-gal activity assay//Cell morphological analysis//qRT-PCR
Description: Cells expressing activated AKT showed a significant increase in SAβGAL and cell size.AKT-induced senescence is characterised by an SASP, with increased secretion of IL-1a, IL-1b, IL-6 and IL-8.

Regulatory relationship

Regulatory pathway: P53//MTORC1
R-AG-Pathway: Upregulation//--
Official symbol(s): TP53//NULL
Pathway experiment: Knockdown//SA-β-gal activity assay//Western blot
Pathway description: SAβGAL positivity was significantly reduced in BJ-T-myr-AKT/p53 stable knockdown cells as compared with control BJ-T-myr-AKT cells. Acute p53 knockdown, confirmed by immunoblot analysis, rescued the reduced proliferation of myr-AKT1-expressing cells to that of control cells .Upon treatment with rapamycin, the percentage of AKT cells positive for SAβGAL was significantly reduced .Rapamycin treatment also dramatically reduced AKT-induced effects on cell size, and the SASP, indicating that mTORC1 activity is critical for PI3K/AKT-driven senescence.
