Description: We depleted HMGB1 in human fibroblasts (IMR-90, HCA2) that reduced intracellular HMGB1 protein levels by >90%. HMGB1-depleted cells underwent senescence, as judged by arrested growth, senescence-associated heterochromatin foci (SAHF) formation (Narita et al., 2003), and SA-β-Gal activity.HMGB1 depletion also caused senescence in human mammary epithelial cells (HMECs) and MEFs, indicating the response was not restricted to human fibroblasts.
