Description: | HFF1 but not HFF3 cells showed intense staining for SA-β-Gal at PDL 50.There was a significant increase in the percentage of SA-β-Gal-positive HFF1 cells upon serial passage. In comparison, such increase was modest for HFF3 cells .Here we showed that the intracellular NADPH/NADP+ ratio of HFF1 cells decreased by around 50% as they were passaged from PDL 12 to 50.In contrast, the NADPH/ NADP+ ratio of HFF3 cells, which was about 50% higher than that of HFF1 at PDL 12, diminished only slightly. The change in NADPH/NADP+ ratio paralleled that of intracellular G6PD activity.The G6PD activity in HFF1 cells decreased significantly with the increasing PDL, whereas that of HFF3 cells was only slightly reduced. The level of 8-OHdG, an oxidative DNA damage marker, in HFF1 cells increased significantly with PDL. |
