Description: | Results showed a greater number of positively stained CD40L-WT/CD40L-M cells than control cells in A549/TR, A549/DDP, and H460 cells.We then explored the effects of CD40L-WT/CD40L-M overexpression on DNA synthesis and cell proliferation, which are characteristics of cell senescence. The results showed that overexpression of CD40L-WT/CD40L-M inhibited DNA synthesis and cell proliferation. |

Pathway description: | CD40L-WT or CD40L-M-overexpressed cells showed nuclear translocation of NF-κB p65.To address the functional significanof NF-κB in the senescence program of CD40L-M-overexpressing NSCLC cells, transfected cells were treated with the NF-κB inhibitor Bay11-7082 . Results of this experiment showed a decrease in the expression of p53. Further, NF-κB signaling was inhibited via p65 siRNA, and the protein levels of p53 and p21 were attenuated. SA-β-gal staining was also detected. |
