Description: | About 54.2% ( P ?0.001) of MCF7 cells expressing hepaCAM were found to have flat enlarged morphology, a typical characteristic of senescent cells. 40% ( P ?0.001) of MCF7 cells expressing hepaCAM were stained blue representing β-galactosidase activity. Conversely, 12.2 and 15.6% of cells expressing vector or hCAM-tailless showed β-galactosidase staining, respectively. |

Pathway description: | Higher protein levels of p53, p21 and p27, but lower levels of cyclin B1 and cdc2, were detected in cells expressing wild-type hepaCAM when compared with cells expressing vector and hCAM-tailless.MCF7 cells expressing hepaCAM were either untransfected or transfected with either control or p53 siRNA, and the protein levels of p21 and p27 were evaluated. In comparison with either the untransfected cells or cells transfected with control siRNA, knockdown of p53 by p53 siRNA lowered the expression of p21 but did not significantly change p27, indicating that, in the presence of hepaCAM, the increase of p21 expression is p53 dependent. In addition, downregulation of p53 in cells expressing hepaCAM resulted in decreased β-galactosidase staining, reduced cell size and increased cell growth, suggesting that suppression of p53 alleviates senescence induced by hepaCAM. |
