Description: Decreased cell proliferation in Doxo- or IR-treated cells was partially recovered by XAF1 downregulation. Repression of XAF1 levels in senescent cells caused morphological changes similar to young cells and a decrease in SA-β-gal activity. Additionally, XAF1 downregulation in premature senescent cells decreased the population of cells in the G1 phase and increased the population of cells in the S and G2/M phases.

Pathway description: The Doxo- and IR-induced increase in p53 expression and activation was reduced in the XAF1 knockdown cells .By measuring the SA-β-gal activity, we demonstrated that p53 knockdown inhibited XAF1-induced cellular senescence but p16 knockdown did not.
