Description: Doxycycline-induced H-RASV12?expression induces cellular senescence in normal BJ human fibroblasts.Few cells stained positive for (SA-β-GAL) activity during the proliferative phase at day 4 but SA-β-GAL positivity increased at day 12 in DOX-treated but not in untreated cultures, coinciding with the onset of growth arrest. By day 16 around 65 % of DOX-treated cells were SA-β-GAL positive compared to 15% of the control cells.?DOX stimulation increased the percentage of cells that showed intense H3K9me3 staining (90 % compared to 30 % in the control), appearing as SAHF or a more widespread nuclear staining .

Target gene description: RAS-induced SA-β-GAL activity was almost completely abolished in p53 knockdown cells, which is in good agreement with their continuous proliferation. Further, only a modest increase in the expression of p16 and H3K9me3 was observed in DOX-treated shp53 cells compared with shCtrl cells, suggesting that p53 depletion inhibited senescence induced by RAS.
