Cell name: | U87,U251 |
Experiment: | Knockdown//SA-β-gal activity assay |
Description: | U251 and U87 cell growth was inhibited by CPEB1 knockdown. Senescent cells can be detected with an assay that detects senescence-associated β-gal activity. |
Target gene: | MIR-101//P53 |
Official symbol(s): | MIR-101//P53 |

Target gene experiment: | Luciferase reporter assay//qPCR |
Target gene description: | When the cells were individually cotransfected with miR-101 and pMIR-CPEB1-30-UTR-WT, there was a significant reduction in luciferase activity compared with cells transfected with the negative control, thereby confirming that miR-101 directly interacted with the 30-UTR of CPEB1 in glioma cells. It was gratifying to observe that miR101 could inhibit the expression of CPEB1, and that the suppression of miR-101 could enhance the expression of CPEB1.After the knockdown of CPEB1, the level of expression of p53 was decreased in the U251 cells but not changed in the U87 cells. Furthermore, it was discovered that the knockdown of CPEB1 induced the overexpression of p21, which is downstream of p53. |
