Description: The efficiency of knockdown was confirmed with mRNA and protein level analyses. We then explored the effects of COX5B depletion on cell proliferation and migration. The result showed that loss of COX5B inhibited both cell proliferation and migration in the three cell lines. In the process, cells with sh-COX5B became flattened and elongated after three passages, which were morphological transformation of cell senescence. Thus, a β-galactosidase staining assay was used to determine whether the cells undergo cell senescence. We found that more COX5B-depleted cells (25~45%) stained positively than control cells (7~16%), suggesting that loss of COX5B induced senescence.
