Description: | Cell cycle analysis revealed prolonged exposure to TNFα significantly impaired proliferation of cells, as evidenced by progressive decline in abundance of BrdU-incorporating cells accompanied by a decreased percentage of Sphase and increased G1/G2-phase cells compared to control, indicating proliferative arrest.Cells exposed to TNFα also developed classic senescent phenotypes, including a flattened, multi-nucleated, giant cell morphology, and concomitantly increased expression of senescence markers, including p16, p21, and cellular SA-β-gal activity. |
