Aging Overview

Aging gene

Gene name: TNF
Aging type: Accelerate
Aging characteristic:
Tissue type: --
Cell name: HUVEC
Gene ID: 7124
Category: protein coding
Phenotype: Aging
Experimental category: HL
PMID: 29176033
Experiment: Flow cytometry//BrdU assay//Western blot//SA-β-gal activity assay
Description: Cell cycle analysis revealed prolonged exposure to TNFα significantly impaired proliferation of cells, as evidenced by progressive decline in abundance of BrdU-incorporating cells accompanied by a decreased percentage of Sphase and increased G1/G2-phase cells compared to control, indicating proliferative arrest.Cells exposed to TNFα also developed classic senescent phenotypes, including a flattened, multi-nucleated, giant cell morphology, and concomitantly increased expression of senescence markers, including p16, p21, and cellular SA-β-gal activity.

Regulatory relationship

Regulatory pathway: JAK-STAT
R-AG-Pathway: Activation
Official symbol(s): hop-SOAT1
Pathway experiment: SA-β-gal activity assay//Western blot
Pathway description: The abundance of serine-phosphorylated STAT1 in cells undergoing senescence due to TNFα was sustained at day 3 and remained elevated at day 6, concomitant with the establishment of senescence .The phosphorylated forms of STAT1 and STAT3 peaked early and then slowly decayed in cells treated with conditioned medium compared to control.
