Description: Our results indicated that the overexpression of SIRT3 significantly reduced the number of cells positive for SA-β-gal and inhibited SAHF formation compared with empty vector .Meanwhile, the accumulation of p16INK4A was decreased .

Pathway description: We found that the endogenous SIRT3 was highly expressed in the mitochondria and cytoplasm, FOXO1 was expressed in the cytoplasm and nuclei, and the SIRT3 staining strongly overlapped with that of FOXO1 in the cytoplasm. We could confirm that SIRT3 was binding with FOXO1. SIRT3 overexpression keeps FOXO1 acetylation to a very low level.To verify the direct effect of SIRT3 on FOXO1-targeted gene expression including catalase and MnSOD, we examined the expression of target genes. The results indicated that expression levels were increased in the pSIRT3 group compared with control and vector groups.
