Gene name: | SIRT1 |
Aging type: | Prevent |
Aging characteristic: |
Tissue type: | -- |
Cell name: | 2BS |
Experiment: | SA-β-gal activity assay//Immunostaining |
Description: | 2BS with enforced expression of wild type SIRT1 displayed lower frequency of SA-β-gal staining than cells transfected with mock vector and H363YSIRT1 constructs. We observed that 2BS cells expressing SIRT1 did not develop pronounced nucleoli or DNA foci. |
Regulatory pathway: | P16-RB//ERK-S6K1 |
R-AG-Pathway: | --//Activation |
Pathway experiment: | Western blot |
Pathway description: | Compared with mock vector and H363YSIRT1 transfected 2BS cells, p16INK4A displayed a two-fold decrease in SIRT1-transfected cells, accordingly phospho-Rb (Ser795) was increased for more than two times whereas the levels of b-actin and Rb remained unchanged in different batches of transfected cells. The levels of SIRT1 protein were declined in late passage of 2BS (63PDs), and the phosphorylations of ERK and S6K1, but not that of AKT, were also decreased in senescent cells. |
