We found that human recombinant Sirt1 bound to caveolin 1 (residues 82–101)-GST.The incubation with the purified GST-Cav-1 (82–101) peptide inhibited Sirt1 activity by- 45% compared to GST alone. |

Pathway description: | We found that human recombinant Sirt1 bound to caveolin 1. However, Sirt1 strongly interacted with caveolin-1 72 hours after stimulation of WI-38 cells with sublethal doses of hydrogen peroxide that we have previously shown induce premature senescence.In contrast, free radicalinduced acetylation of p53 was significantly inhibited in cells lacking caveolin-1. Interestingly, downregulation of Sirt1 by siRNA enhanced oxidant-induced acetylation of p53 in caveolin-1 null MEFs to levels seen in wild type MEFs. |
