Aging Overview

Aging gene

Gene name: SIRT1
Aging type: Prevent
Aging characteristic:
Tissue type: --
Cell name: WI-38
Gene ID: 23411
Category: protein coding
Phenotype: Aging
Experimental category: HL
PMID: 12006491
Experiment: SA-β-gal activity assay//BrdU assay
Description: As observed for the growth curves, expression of SIRT1 alone does not significantly affect the percentage of cells in S phase or the percentage of SA-β-gal-positive cells when compared with control infected cells. However, expression of PML IV results in a striking decrease of BrdU-positive cells and a huge increase in SA-β-gal Activate cells, as reported previously. Double-infected PML IV–SIRT1 cells exhibit a significant increase in the percentage of cells in S phase as compared with PML-infected cells, in addition to a marked decrease in the percentage of SA-β-gal-positive cells.

Regulatory relationship

Target gene: P53
Official symbol(s): P53
R-AG-Target gene: Inhibition
Subcategory: Unclear
Target gene experiment: Pull-down assay
Target gene description: We performed pull-down assays with either full-length GST–p53 or a series of GST–p53 fusions expressing only the N-terminal, middle or C-terminal regions of the protein, and in vitro translated SIRT1. We observed specific binding of SIRT1 to full-length p53 as well as to GST–p53(290–393), while weaker binding was seen with GST–p53(90–290) and no binding occurred with the N-terminal p53 fusion.
