Gene name: | NAMPT |
Aging type: | Prevent |
Aging characteristic: |
Tissue type: | -- |
Cell name: | MEF |
Experiment: | SA-β-gal activity assay |
Description: | Over- expression of NAMPT in MEF cells slows down cellular senescence.Percentage of cells that is positive for SA- β- Gal activity in all Wt- MEF cells was dramatically increased from passage 6, consistent with the timing of CCP in Wt- MEF cells.we measured the expression level of several senescence marker genes. p16INK4a, p19ARF, and p21CIP1 are cell cycle inhibitors and their expression levels increase when cells enter senescence. |
Target gene: | SIRT1 |
Official symbol(s): | SIRT1 |
R-AG-Target gene: | Upregulation |
Subcategory: | Unclear |
Target gene experiment: | qRT-PCR |
Target gene description: | We finally examined whether SIRT1 activity is enhanced in response to NAMPT over- expression. Although SIRT1 has been shown to regulate cellular proliferation and senescence, this function is dependent on the robustness of the intrinsic NAMPT activity and NAD+ content. SIRT1 is also vital in regulating FOXO- mediated activation of antioxidant enzymes including SOD2 and Catalase. As we shown above, NAD+ content in Tg- MEF was higher than that in Wt- MEF cells. SIRT1 activity in Tg- MEF was approximately threefold higher than that in Wt- MEF cells, in line with the observed SOD2 and catalase up regulation. |
